Hexapeptide-9, also known as Collagen Peptide III (Gly‐Pro‐Gln‐Gly‐Pro‐Gln), is one of my favorite topical bio-actives in clinical formulation, and a star ingredient in our Peptide Firming Formulation. Here's why it earns its place.
What Is Hexapeptide-9?
Hexapeptide-9 is a synthetic six-amino-acid peptide that mirrors a fragment found in human collagen IV and XVII, two key structural proteins of the skin's basement membrane. Because it resembles the body's own collagen, it can bind directly to collagen receptors on fibroblasts, the cells in your dermis responsible for producing collagen and elastin.
This binding action sends a clear signal: make more collagen. The result is increased production of collagen and other extracellular matrix (ECM) proteins, including elastin and glycosaminoglycans, which attract and retain moisture leading to measurably firmer, denser, and more resilient skin.
What Else Does Hexapeptide-9 Do?
- Reduces collagen degradation — Its anti-inflammatory and antioxidant properties help protect existing collagen and preserve the skin barrier.
- Accelerates repair and regeneration — It activates key growth factors involved in tissue healing, including TGF-β (Transforming Growth Factor-beta), a powerful protein that supports ECM repair.
- Strengthens the skin barrier — Hexapeptide-9 promotes the production of extracellular lipids such as ceramides and filaggrin (a filament-aggregating protein), reinforcing the adhesiveness of barrier proteins. A stronger barrier means better protection against microbes, allergens, and pollutants and significantly less trans epidermal water loss (TEWL), keeping skin soft and hydrated.
To put the barrier in perspective: it's no thicker than a single sheet of paper, yet it's your skin's first line of defense against everything the environment throws at it. Keeping it intact is essential to the health of the skin.
How I Use It Clinically

Peptides vs. Retinol - or Both?
If you're sensitive to retinol or Retin-A, peptides are an excellent alternative. They're highly effective, exceptionally stable, and cause no irritation making them suitable for even reactive skin types.
That said, if your skin tolerates retinol well, I strongly recommend combining both. Together, peptides and retinol address fine lines, wrinkles, loss of elasticity and volume, skin thinning, sun damage, and the full spectrum of age-related skin concerns more comprehensively than either can alone.
